A new research study by scientists in Australia and the US provides an explanation for why clinical trials of drugs reducing proteins in the brain that were thought to cause dementia and Alzheimer's have failed. The study has opened the way for potential new treatments with existing drugs. Published online in the journal Human Molecular Genetics, the researchers assembled evidence from a wide range of human studies and animal models of dementia-related diseases to show that inflammation is a major cause, not just a consequence. They show that many genes linked with dementia regulate our susceptibility and response to inflammatory damage. " For decades, scientists have thought that dementia and Alzheimer's Disease are caused by protein aggregates forming in the brain. But recent clinical trials of drugs that reduce the aggregates have failed ," says project leader Professor Robert Richards, from the University of Adelaide's School of Biological Sciences....

Merck is investing $125 million in Moderna under a new deal that expands the firms’ 2016 partnership to develop messenger RNA cancer vaccines. The deal now includes shared antigen mRNA cancer vaccines including mRNA-5671, Moderna’s mRNA KRAS cancer vaccine, the firms noted. The two companies will now jointly advance mRNA-5671 in human studies , and combination studies with additional immuno-oncology therapies are also in the pipeline . Merck will be responsible for clinical development of mRNA-5671 and associated costs while Moderna will be responsible for clinical supply and linked costs. On completion of human proof-of-concept studies, Merck may opt-in on further development and commercialization of the vaccine on payment of an undisclosed fee to Moderna. “ Augmentation of immune responses offers great promise in cancer therapy, as our work with the PD-1-specific antibody Keytruda has shown ,” said Dr Roger M. Treatment for the dreaded disease tuberculosis, spreading in alarming proportion in India, has been largely ignored by most drug companies because of "limited or no financial incentive" and lack of basic research, according to Dr Paul Stoffels, executive VP and chief scientific officer of US-based drug biggie Johnson & Johnson (J&J). In another milestone for Indian pharmaceutical major Glenmark Pharmaceuticals, the company has announced positive data from its phase 2a study of GBR 830, an investigational anti-OX40 monoclonal antibody for treating atopic dermatitis. In the double-blind, placebo-controlled study, researchers randomly assigned 62 patients (52% male) with moderate-to-severe atopic dermatitis to measure the safety, biological and clinical activity and prokinetics of GBR 830 over 12 weeks, according to a news release. Thirty-one patients were evaluated on multiple endpoints following the last study visit, after receiving two doses with two viable biopsies. Seventeen out of 23 patients in the GBR 830 cohort experienced at least a 50% reduction in the Eczema Area and Severity Index (EASI) scores at day 57 when compared to baseline, according to the release.
